pMDLg/pRRE质粒慢病毒包装载体简介

第三代慢病毒包装质粒系统包括有:Gag基因作用是编码病毒粒子主要结构蛋白;Pol基因作用是编码特异性的逆转录酶;RRE基因作用是Rev蛋白的结合位点,有利于RNA的出核转运。pMDLG/pRRE质粒不含有Rev基因。第三代慢病毒包装质粒提供了最大的生物安全性,但是由于需要同时转染四个不同的质粒到产毒细胞中,在操作上也更加复杂。

常用的第三代慢病毒包装质粒组合是: pMDLg/pRRE + pRSV-Rev + pMD2.G.

请注意,大部分的Trono实验室慢病毒载体含有一个野生型的5' LTR 。这种类型的慢病毒载体,只能够使用第二代慢病毒包装质粒进行包装。这是由于野生型的5' LTR必须要有TAT序列,才能够被激活。

如果你想使用第三代慢病毒包装质粒,你的慢病毒目的基因质粒必须含有一个嵌合型的5' LTR片段(例如CCL-, RRL-等基因),在该片段中HIV启动子被CMV和RSV启动子取代了,所以该慢病毒质粒,并不依赖于TAT系统。

含有嵌合型5' LTR的慢病毒质粒,既能够使用第二代慢病毒包装质粒,也能够使用第三代慢病毒包装载体。
