pCold IV质粒 pColdIV载体简介
阐明蛋白质的结构和功能是后基因组研究的一个重要课题。高效率的蛋白质表达系统是研究这些课题的不可缺少的基本技术。大肠杆菌作为宿主的表达系统中被广泛 使用于生产重组蛋白。大肠杆菌表达系统具有良好的优势,方法简单,成本低。然而,对于某些基因,蛋白表达很困难,表达的蛋白质不溶。
为了解决这些问题,TAKARA与Masayori Inouye教授(美国新泽西医学和牙科大学)进行联合研究,以制定一个高效基于低温冷休克蛋白基因的大肠杆菌表达载体。pCold DNA系列产品具有上述的优点,并提供了一个重要功能和结构的分析以及其他蛋白质研究领域的工具。
<产品概况和特点>
当培养的大肠杆菌的温度充分降低,几乎暂时停止生长大部分的蛋白表达减少,而此时一组蛋白质称为“冷休克蛋白”被专门诱导表达。
冷休克表达载体,pCold I-IV,是利用冷休克基因CSPA启动子设计高效率蛋白质表达载体。 CSPA的启动子的下游处,插入了lac操纵子,以便严格控制表达。此外,5'非翻译区(5'UTR),翻译增强元件(TEE),His标签序列,Xa因 子切割位点和多克隆位点(MCS)位于在CSPA启动子的下游的。由于该产品采用了大肠杆菌的启动子,大多数大肠杆菌菌株可被用作表达宿主。有四种类型的 pCold载体,他们的下列原件的位置稍有不同,TEE,His标签序列和Factor XA酶切位点。
