pRSET B质粒 pRSETB载体简介
pRSET B载体是从pUC的载体衍生而来,载体设计的目的是为了在大肠杆菌中高水平的表达和纯化目的克隆基因。载体上的T7启动子使得pRSETB载体高水平表达克隆的基因序列成为可能。
此外,DNA插入片段的位于载体上面的下游侧,并能够上游的蛋白编码框相一致,上游的编码序列表达了一个N-端融合的肽。该序列包括一个ATG翻译起始密 码子,一个6 X His标签,一个来源于噬菌体T7基因10的转录稳定序列,Xpress抗原表位,以及一个肠激酶裂解识别序列。
His 标签允许简单的Ni柱蛋白纯化。肠激酶识别位点位于His标签和重组蛋白之间,可以随后方便的去除重组蛋白的N-端的融合肽。
