pUG66载体是大肠杆菌-酵母穿梭载体,在大肠杆菌中抗性是氨苄。pUG66载体质粒含有来自Tn5载体质粒的Ble基因,使得载体质粒在酵母菌中呈现出博来霉素抗性。 pUG66载体在筛选基因Ble两侧含有loxp基因,通过表达cre重组酶,可以利用loxp基因将Ble s筛选基因删除。 pUG66载体可以作为酵母基因敲除的一个质粒PCR模板。代表性的PCR片段的整合插入宿主菌,使得基因敲除突变子能够进行顺利筛选。 |
